Protein-based mechanism of wheat growth under salt stress in seeds irradiated with millimeter waves. 招待査読国際誌

Wheat is one of the most extensively grown crops in the world; however, its productivity is reduced due to salinity. This study focused on millimeter wave (MMW) irradiation to clarify the salt-stress tolerance mechanism in wheat. In the present study, wheat-root growth, which was suppressed to 77.6% of the control level under salt stress, was recovered to the control level by MMW irradiation. To reveal the salt-stress tolerance mechanism of MMW irradiation on wheat, a proteomic analysis was conducted. The results indicate that MMW irradiation of wheat seeds improves plant growth recovery from salt stress through regulating the reactive oxygen species scavenging system and the pathogen-related proteins. These genes may contribute to the development of salt-stress-tolerant wheat through marker-assisted breeding and genome editing.

Finding solutions to mitigate the impact of salinity on crops is important for global food security because soil salinity significantly reduces plant growth and grain yield. Ethanol may play an important role in mitigating the negative salt-induced effects on crops. Soybean root growth was significantly reduced under salt stress; however, it was restored and comparable to control values by ethanol application even under stress. To study the positive mechanism of ethanol on soybean growth, a proteomic approach was carried out. The results suggest that soybeans were adversely affected by salt stress and recovered with ethanol application via the regulation of cell wall and membrane functions through the detoxification of reactive oxygen species.

Salt stress is a serious problem, because it reduces the plant growth and seed yield of wheat. To investigate the salt-tolerant mechanism of wheat caused by plant-derived smoke (PDS) solution, metabolomic and proteomic techniques were used. PDS solution, which repairs the growth inhibition of wheat under salt stress, contains metabolites related to flavonoid biosynthesis. The results suggest that PDS solution enhances wheat growth suppressed by salt stress through the regulation of energy metabolism and the ubiquitin-proteasome system related to flavonoid metabolism.

The establishment of soybean seedlings is sensitive to flooding, and colocalization of proteins and quantitative trait loci (QTLs) advances to narrow down the hub genes. To reveal soybean seedling tolerance to flooding, Qihuang34 (QH34), Jidou17(JD17), and their recombinant inbred lines were used in this study. The results suggest that integrating proteomics with QTL is advantageous to identify genes associated with flood tolerance, proposing that enhanced protein homeostasis and cell wall stiffening in root tips may confer soybean seedling tolerance to flooding via GA and JA signals.

Floods induce hypoxic stress and reduce wheat growth. On the other hand, rice is a semi-aquatic plant and usually grows even when partially submerged. To clarify the dynamic differences in the cellular mechanism between rice and wheat under flooding stress, morphological and biochemical analyses were performed. The results suggest that flooding stress significantly inhibits wheat growth through upregulating amino-acid metabolism and increasing the alcohol-fermentation system compared to rice. When plant growth is inhibited by flooding stress and the aerobic-metabolic system is activated, GABA content increases.

The irradiation with millimeter-wave (MMW) of wheat seeds promotes root growth under flooding stress; however, its role is not completely clarified. Nuclear proteomics was performed, to reveal the role of MMW irradiation in enhancing flooding tolerance. The purity of nuclear fractions purified from roots was verified. Histone, which is a protein marker for nuclear-purification efficiency, was enriched; and cytosolic ascorbate peroxidase was reduced in the nuclear fraction. The results suggest that MMW irradiation improves wheat-root growth under flooding stress through the regulation of mRNA-expression level and the ubiquitin-proteasome system.

Plant-derived smoke is known as a potent agent to promote plant growth at different developmental stages. The application ofplant-derived smoke elicits germination and post-germination stages of plant species. On the other hand, abiotic stresses, such as salinity, heavy metals, and flooding, are known to weaken plant growth and development. Abiotic stresses have negative effects on growth attributes of crops, such as maize, rice, wheat, and soybean. The positive/alleviating effect of plant-derived smoke solution was observed at morphological, physiological, and molecular levels of plant under abiotic stresses. The role of plant-derived smoke in alleviation of various stresses has been comprehended and the mechanism of alleviation via regulation of cellular activities at molecular level has been discussed.

Safranal is a free radical scavenger and useful as an antioxidant molecule; however, its promotive role in soybean is not explored. Salt stress decreased soybean growth and safranal improved it even if under salt stress. To study the positive mechanism of safranal on soybean growth, a proteomic approach was used. The results suggest that safranal improves soybean growth through the regulation of cell wall and nuclear proteins along with reactive oxygen species scavenging system. Furthermore, it might promote salt-stress tolerance through the regulation of membrane proteins involved in endocytosis and post-Golgi trafficking.

Rice plants silenced for peroxisomal ascorbate peroxidase display higher resilience to photosynthesis under oxidative stress and photorespiratory conditions. However, the redox mechanisms underlying that intriguing response remain unknown. The hypothesis was tested that favorable effects triggered by peroxisomal APX deficiency on photosynthesis resilience under CAT inhibition are dependent on the intensity of photorespiration associated with the abundance of photosynthetic and redox proteins. The results suggest that the intensity of photorespiration and peroxisomal APX-mediated redox signaling are tightly regulated under CAT inhibition induced oxidative stress, which can modulate the photosynthetic efficiency, possibly via a coordinated regulation of protein abundance and rearrangement, ultimately triggered by crosstalk involving H2O2 levels related to CAT and APX activities in peroxisomes.

Flooding induces low-oxygen stress, which reduces plant growth. The activity of the γ-aminobutyric acid (GABA) shunt is crucial for plant stress adaptation, in which it acts by changing cytosolic pH, limiting reactive oxygen species production, regulating nitrogen metabolism, and bypassing steps in the tricarboxylic acid cycle. GABA accumulates under osmotic stress as well as flooding stress. To clarify the dynamic roles of GABA accumulation in wheat under flooding stress, biochemical and enzymatic analyses were performed using a plant-derived smoke solution (PDSS), which rescued wheat growth from flooding stress. The results suggest that flooding stress increases GABA content along with the increase and decrease of glutamate decarboxylase and succinyl semialdehyde dehydrogenase, respectively. Additionally, PDSS increased GABA content along with the increase of glutamate decarboxylase abundance at the initial stage of application.

Higher production of secondary metabolites is one of the adaptive responses to alleviate the impact of environmental injuries. In the present investigation, the production of these metabolites with medicinal importance induced by salinity in Carum copticum was investigated. To develop a better way for the production of medicinal substances, callogenesis and plant regeneration were analyzed, and seeds, calli, and/or regenerated seedlings were exposed to different concentrations of NaCl under in vitro culture conditions. The maximum frequency of callus induction was obtained on a medium supplemented with 0.25 mg L􀀀1 2, 4-dichlorophenoxyacetic (2,4-D) and 1 mg L􀀀1 benzyl amino purine (BAP) from stem explants. Plant regeneration with multiple shoots was obtained from pieces of callus transferred to MS medium fortified with 0.25 mg L􀀀1 2,4-D and 1.5 mg L􀀀1 BAP. Four weeks after treatment, salinity induced a substantial increase in the accumulation of reducing sugars and proline as compatible osmolytes and the activity of antioxidant enzymes. Total phenolics and anthocyanin significantly increased in all samples with increasing NaCl concentrations; however, the regenerated seedlings showed a reduction in these compounds at severe NaCl concentration compared to the control. Moreover, NaCl enhanced thymol, -terpinene, sabinene, and myrcene in the seedlings and calli, as well as carvacrol, limonene, and _-terpinene in the regenerated seedlings. These results suggest that salinity has a marked impact on improving the content of antioxidant metabolites and essential oils in C. copticum, whose callus might be the most salt tolerant in all tested samples.

Plant-mediated synthesis methods of nanoflowers (NFs) open a new epoch of extensive application, economical, exceedingly stable, and reproducible synthesis of zinc oxide (ZnO) NFs. To study the effects of bio-synthesized (BS) NFs and chemically synthesized nanowires (CS NWs) ZnO on soybean, a gel-free/ label-free proteomic technique was used. Length and weight of root including hypocotyl were enhanced by 10 ppm BS NFs and CS NWs ZnO. Oppositely changed proteins between BS NFs and CS NWs ZnO were related to hormone metabolism, protein folding, and redox metabolism. Abundance of heat shock protein 70 (HSP70) increased in BS NFs ZnO while did not change in CS NWs ZnO. Abundance of ascorbate peroxidase and peroxiredoxin increased and decreased, respectively, in BS NFs ZnO. Ascorbic acid and hydrogen peroxide increased, respectively, with BS NFs and CS NWs ZnO. These results suggest that BS NFs ZnO might improve the soybean growth by increasing the protein folding due to accumulation of HSP70 and redox metabolism through detoxification of hydrogen peroxide. On the other hand, CS NWs ZnO treated soybean experienced loss of protein folding and increased oxidative stress.

Flooding impairs wheat growth and considerably affects yield productivity worldwide. On the other hand, irradiation with millimeter waves enhanced the growth of chickpea and soybean under flooding stress. In the current work, millimeter-wave irradiation notably enhanced wheat growth, even under flooding stress. To explore the protective mechanisms of millimeter-wave irradiation on wheat under flooding, quantitative proteomics was performed. According to functional categorization, proteins whose abundances were changed significantly with and without irradiation under flooding stress were correlated to glycolysis, reactive-oxygen species scavenging, cell organization, and hormonal metabolism. Immunoblot analysis confirmed that fructose-bisphosphate aldolase and _ tubulin accumulated in root and leaf under flooding; however, even in such condition, their accumulations were recovered to the control level in irradiated wheat. The abundance of ascorbate peroxidase increased in leaf under flooding and recovered to the control level in irradiated wheat. Because the abundance of auxin-related proteins changed with millimeter-wave irradiation, auxin was applied to wheat under flooding, resulting in the application of auxin improving its growth, even in such condition. These results suggest that millimeter-wave irradiation on wheat seeds improves the recovery of plant growth from flooding via the regulation of glycolysis, reactive-oxygen species scavenging, and cell organization. Additionally, millimeter-wave irradiation could promote tolerance against flooding through the regulation of auxin contents in wheat.

Morphological and proteomic analyses of soybean seedling interaction mechanism affected by fiber crosslinked with zinc-oxide nanoparticles. 招待査読国際誌

Nanoparticles (NPs) enhance soybean growth; however, their precise mechanism is not clearly understood. To develop a more effective method using NPs for the enhancement of soybean growth, fiber crosslinked with zinc oxide (ZnO) NPs was prepared. The solution of ZnO NPs with 200 nm promoted soybean growth at the concentration of 10 ppm, while fibers crosslinked with ZnO NPs promoted growth at a 1 ppm concentration. Soybeans grown on fiber cross-linked with ZnO NPs had higher Zn content in their roots than those grown in ZnO NPs solution. To study the positive mechanism of fiber crosslinked with ZnO NPs on soybean growth, a proteomic technique was used. Proteins categorized in photosynthesis and secondary metabolism accumulated more in soybeans grown on fiber crosslinked with ZnO NPs than in those grown in ZnO NPs solution. Furthermore, significantly accumulated proteins, which were NADPH oxidoreductase and tubulins, were confirmed using immunoblot analysis. The abundance of NADPH oxidoreductase increased in soybean by ZnO NPs application. These results suggest that fiber crosslinked with ZnO NPs enhances soybean growth through the increase of photosynthesis and secondary metabolism. Additionally, the accumulation of NADPH oxidoreductase might relate to the effect of auxin with fiber crosslinked with ZnO NPs on soybean growth.

Wheat is an important staple food crop for one‐third of the global population; however, its growth is reduced by flooding. On the other hand, a plant‐derived smoke solution enhances plant growth; however, its mechanism is not fully understood. To reveal the effects of the plant‐derived smoke solution on wheat under flooding, morphological, biochemical, and proteomic analyses were conducted. The plant‐derived smoke solution improved wheat‐leaf growth, even under flooding. According to the functional categorization of proteomic results, oppositely changed proteins were correlated with photosynthesis, glycolysis, biotic stress, and amino‐acid metabolism with or without the plant‐derived smoke solution under flooding. Immunoblot analysis confirmed that RuBisCO activase and RuBisCO large/small subunits, which decreased under flooding, were recovered by the application of the plant‐derived smoke solution. Furthermore, the contents of chlorophylls a and b significantly decreased by flooding stress; however, they were recovered by the application of the plant‐derived smoke solution. In glycolysis, fructose‐bisphosphate aldolase and glyceraldehyde‐3‐phosphate dehydrogenase decreased with the application of the plant‐derived smoke solution under flooding as compared with flooding alone. Additionally, glutamine, glutamic acid, aspartic acid, and serine decreased under flooding; however, they were recovered by the plant derived smoke solution. These results suggest that the application of the plant‐derived smoke solution improves the recovery of wheat growth through the regulation of photosynthesis and glycolysis even under flooding conditions. Furthermore, the plant‐derived smoke solution might promote wheat tolerance against flooding stress through the regulation of amino‐acid metabolism.

Proteomic, biochemical, and morphological analyses of the effect of silver nanoparticles mixed with organic and inorganic chemicals on wheat growth 招待査読国際誌

Wheat is vulnerable to numerous diseases; on the other hand, silver nanoparticles (AgNPs) exhibit a sterilizing action. To understand the combined effects of AgNPs with nicotinate and potassium nitrate (KNO3) for plant growth and sterilization, a gel- and label-free proteomics was performed. Root weight was promoted by the treatment of AgNPs mixed with nicotinate and KNO3. From a total of 5557 detected proteins, 90 proteins were changed by the mixture of AgNPs, nicotinate, and KNO3; among them, 25 and 65 proteins increased and decreased, respectively. The changed proteins were mainly associated with redox and biotic stress in the functional categorization. By immunoblot analysis, the abundance of glutathione reductase/peroxiredoxin and pathogen-related protein three significantly decreased with the mixture. Furthermore, from the changed proteins, the abundance of starch synthase and lipoxygenase significantly increased and decreased, respectively. Through biochemical analysis, the starch contents increased with the mixture. The application of esculetin, which is a lipoxygenase inhibitor, increased the weight and length of the root. These results suggest that the AgNPs mixed with nicotinate and KNO3 cause positive effects on wheat seedlings by regulating pathogen-related protein and reactive-oxygen species scavenging. Furthermore, increasing starch and decreasing lipoxygenase might improve wheat growth.
